Rationale: Chronic Daily Headache (CDH) significantly reduces quality of life, and current management strategies often fail to provide consistent relief. Magnesium, an essential micronutrient involved in neurological function, has been suggested to play a role in headache pathophysiology, and additionally, genetic variations in the fat mass and obesity-associated (FTO) gene may influence individual responses to dietary factors. This study aimed to investigate the association between dietary magnesium intake and FTO gene polymorphisms  in individuals with CDH.

 

Methods: This case-control study included 150 patients with CDH and 150 control participants aged 40 to 80 years. Magnesium intake was assessed using a validated food frequency questionnaire (FFQ). The FTO rs9939609 polymorphism was genotyped using the tetra-primer amplification refractory mutation system polymerase chain reaction (ARMS-PCR).

 

Results: Dietary magnesium intake was marginally lower among CDH cases compared to controls within the AT/AA genotype group; however, this difference was not statistically significant. Logistic regression analysis identified a significant inverse association between dietary magnesium intake and CDH risk in individuals with the AT/AA genotype (OR = 0.995; 95% CI: 0.990–1.000; p = 0.040). No significant association was observed in the TT genotype group.

Conclusion: This study demonstrates a significant interaction between dietary magnesium intake and genetic susceptibility in the development of CDH. Further prospective studies are required to validate these findings.
