TGF-Β2 DETECTION IN A SEMI-ELEMENTAL FORMULA FOR NUTRITIONAL SUPPORT IN MALABSORPTION

Rationale: The presence of TGF-β2 in nutritional formulas may offer immunomodulatory and regenerative benefits for patients with gastrointestinal disorders such as malabsorption. This study aims to quantify the concentration of TGF-β2 in a semi-elemental formula based on hydrolyzed whey protein.

Methods: The quantification of bovine TGF-β2 was performed using a sandwich-type ELISA assay. The methodology was based on the immobilization of anti-TGF-β2 antibodies, followed by the addition of samples and biotinylated detection antibodies. After incubation, washing, and addition of HRP-Streptavidin, the reaction was visualized with TMB substrate and stopped with a stop solution. The absorbance was read at 450 nm, and the concentration was determined using a standard curve fitted with a four-parameter logistic (4PL) model, with R² = 0.992. Samples of the formula (Peptimax – Prodiet Medical Nutrition) were analyzed under two conditions: (1) supernatant without dialysis and (2) dialyzed with a <3.5 kDa cutoff membrane to remove low molecular weight interferents.

Results: The concentration of TGF-β2 in the analyzed formula increased from 61.12 pg/mL to 87.70 pg/mL after dialysis with a <3.5 kDa cutoff membrane. This increase indicates that the original matrix contained low molecular weight interferents that inhibited cytokine detection. The removal of these compounds improved assay sensitivity, suggesting that a fraction of TGF-β2 was partially masked.

Conclusion: The semi-elemental formula contains detectable levels of TGF-β2, with concentrations varying according to sample processing methods. Notably, dialysis using a <3.5 kDa membrane increased the measured concentration, underscoring the critical role of sample preparation in accurate quantification. Given the known ability of TGF-β2 to mitigate chemotherapy-induced diarrhoea and mucositis, its presence in Peptimax suggests potential protective benefits for intestinal health.
