EFFECTS OF SYNBIOTIC BASED PRODUCT ON GUT MICROBIOTA AND OVERALL WELLBEING IN ADULTS

Rationale: Gut microbiome dysbiosis is one of the hallmarks of unhealthy aging. This study aimed to investigate health benefits of a unique synbiotic combination incorporated in healthy adults.

Methods: This was a single-arm, self-controlled trial conducted in China (NCT06646458), enrolling 30 participants aged 50-70 years. Participants consumed two daily servings of the milk powder, for 28 consecutive days. Stool samples were collected at baseline (Day 0, V1), Day 7 (V2), Day 14 (V3), and Day 28 (V4).

Results: There were significant increases in total Bifidobacterium abundance, in particular B. longum and B. pseudocatenulatum, at V2, V3, and V4 compared with V1 (all P < 0.05).  In addition, the abundance of several short-chain fatty acid (SCFA)-producing bacteria also increased markedly after intervention. Total SCFAs were all significantly increased compared with baseline (all P < 0.05). Several key microbial metabolic pathways were enriched, including the phosphotransferase system, fructose and mannose metabolism, biosynthesis of secondary metabolites, and amino acid biosynthesis. Most participants (86%) reported marked improvements in quality of life. We observed a positive trend of improved gastrointestinal function and benefits, as revealed by better defecation satisfaction and higher proportion of normal stool consistency after intervention.

Conclusion: Following intervention, there was a significant increase in the abundances of Bifidobacterium species and SCFA-producing bacteria, contributing to increased SCFA production which contributed to overall improvement in health.
