Rationale: The objective was to explore the effects of shelf-stable yogurt containing live LGG on gastrointestinal symptoms, defecation frequency, and stool characteristics in adults aged 45-70.
Methods: In this randomized, double-blind, controlled trial, 76 participants aged 45 to 70 years with FGID-related symptoms were assigned to consume either two packs of shelf-stable fermented yogurt containing live LGG (1×109 CFU per pack) or two packs of conventional shelf-stable fermented yogurt without active probiotics for 8 weeks (n=43 vs. n=33). GSRS scores, self-reported defecation frequency, and BSS scores were assessed at baseline and at weeks 2, 4, and 8, and analyzed using linear mixed-effects models under the intention-to-treat principle.
Results: A significant group × time interaction was observed for GSRS total score (P = 0.022), indicating different temporal trajectories between groups. The constipation syndrome score also showed a significant group × time interaction (P < 0.001), with significant between-group differences at baseline (ΔEMM = 0.53, 95% CI: 0.22 to 0.85, PFDR = 0.001) and week 8 (ΔEMM = -0.60, 95% CI: -0.92 to -0.27, PFDR < 0.001). Significant main effects of time were observed for defecation frequency and BSS scores: defecation frequency increased in participants with constipation at baseline and decreased in those with diarrhea at baseline (both P < 0.001), while BSS scores increased in participants with hard stools at baseline (P = 0.015) and decreased in those with loose stools at baseline (P < 0.001), after adjustment for baseline values and sex.
Conclusion: Shelf-stable yogurt containing live LGG improved GSRS total scores and showed greater benefits for constipation-related symptoms among adults aged 45 to 70 years with FGID-related symptoms. Both yogurts improved defecation frequency and stool characteristics over time.
