Rationale: To determine the effect of fecal microbiota transplantation (FMT) from stunted (S) children on crown-like structure (CLS) density in visceral adipose tissue (VAT) of C57BL/6 mice fed a high-fat, high-fructose corn syrup diet (HFFr).
Methods: FMT used pooled stools from healthy (C) or S school-aged children. Eight-week-old male C57BL/6 mice underwent bowel cleansing with polyethylene glycol, followed by weekly intragastric FMT for four weeks, then fed a control (Ct) or HFFr diet for 15 weeks (groups: C-Ct, C-HFFr, S-Ct, S-HFFr). VAT sections were stained with H&E; CLS/mm² was quantified using ImageJ. Mann-Whitney U tests with Dunn-Bonferroni correction were applied; effect size (r) and superiority percentage complemented p-values.
Results: S-HFFr showed the highest median CLS/mm²; C-Ct and C-HFFr were similar; S-Ct was lowest. Significant differences were found for C-Ct vs. S-HFFr (p = 0.041), C-HFFr vs. S-HFFr (p = 0.026), and S-Ct vs. S-HFFr (p = 0.017); after adjustment, only S-Ct vs. S-HFFr remained significant (p = 0.011). Nevertheless, large Cohen's r effect sizes and superiority percentages exceeding 85% were observed in C-Ct vs. S-HFFr (r = 0.60; 86%), C-HFFr vs. S-HFFr (r = 0.63; 88%), S-Ct vs. S-HFFr (r = 0.69; 91.7%), underscoring a substantial increase in CLS density driven by stunting-associated microbiota and the HFFr diet.
Conclusion: Stunting-associated fecal microbiota combined with a HFFr diet displayed higher VAT CLS density, indicating chronic low-grade adipose inflammation.
