BROCCOLI CONSUMPTION IS ASSOCIATED WITH LOWER RISK OF SARCOPENIA AND FAVORABLE PROTEOMIC PROFILES: A CROSS-SECTIONAL STUDY FROM THE UK BIOBANK

Rationale: Sarcopenia, the progressive loss of muscle, is a major health concern. Broccoli is rich in bioactive compounds, but its link to sarcopenia in large populations is unconfirmed.

Methods: We analyzed 192,193 UK Biobank participants. Broccoli intake (none, <1, ≥1 portion/day) was assessed via dietary recalls. Sarcopenia was defined by low grip strength and low appendicular lean mass (EWGSOP2). Logistic regression models adjusted for confounders. Olink proteomics, LASSO, and mediation analyses explored mechanisms.

Results: Sarcopenia prevalence was 0.47%. Broccoli consumers had lower odds of sarcopenia versus non-consumers (fully-adjusted OR=0.84, 95% CI: 0.71–0.98). A dose-response was observed: intake ≥1 portion/day linked to 24% lower odds (OR=0.76, 95% CI: 0.62–0.92). Proteomics identified 681 differentially expressed proteins in consumers, enriched in immune and extracellular matrix pathways. Mediation analysis nominated 41 potential protein mediators (e.g., FGF21, PCSK9). Broccoli intake also correlated with favorable future body composition.

Conclusion: Broccoli consumption was associated with lower sarcopenia odds in a dose-dependent manner, potentially mediated by inflammation- and tissue remodeling-related proteins. This supports further investigation of cruciferous vegetables for muscle health.
