Rationale: Hot flushes and night sweats are hallmark symptoms during menopause. This post‑hoc analysis addresses whether a probiotic formulation (Gyntima® Menopause) shown to modulate the estrobolome, reduces the frequency and severity of hot flushes in peri‑ and postmenopausal women.
Methods: Randomized, double-blind, placebo-controlled trial. Women in perimenopause (n=95) or postmenopause (n=97) with menopausal symptoms (Cervantes Scale score >38) were recruited. Participants were randomized (2:1) to receive either a probiotic formula comprising Levilactobacillus brevis KABP052, Lactiplantibacillus plantarum KABP051, Pediococcus acidilactici KABP021; 1×10⁹ CFU/day (Gyntima® Menopause) or placebo (1 capsule/day) for 3 months. Changes in the severity of hot flushes were evaluated through the Menopause Rating Scale (MRS-II), item I (Hot flushes, sweating, episodes of sweating). Mixed model for repeated measures and chi-squared tests were used to compare average scores and percent of women with symptom worsening between groups, respectively.
Results: In the group of perimenopausal women the hot flushes score decreased both in active and placebo groups though there were no differences between them. In postmenopausal women, probiotic significantly decreased the average hot flushes score across the study in comparison to placebo (p<0.05, adjusted for baseline). In this group, the percentage of women experiencing a symptom worsening was markedly lower in the probiotic group in comparison to placebo at month 1 (10.0% vs 32.4%, p<0.01) month 2 (5.0% vs 24.3%, p< 0.01) and month 3 (3.3% vs 16.2%, p<0.05).
Conclusion: In this post-hoc analysis, supplementation with the tested probiotic formula (Gyntima® Menopause) modestly improved hot flushes measured by the MRS-II scale in postmenopausal women, especially preventing their worsening.
